We aimed to evaluate the relationship between imaging features, therapeutic responses (comparative cross-product and volumetric measurements), and overall survival (OS) in pediatric diffuse intrinsic pontine glioma (DIPG). A total of 134 patients (≤ 18 years) diagnosed with DIPG were included. Univariate and multivariate analyses were performed to evaluate correlations of clinical and imaging features and therapeutic responses with OS. The correlation between cross-product (CP) and volume thresholds in partial response (PR) was evaluated by linear regression. The log-rank test was used to compare OS patients with discordant therapeutic response classifications and those with concordant classifications. In univariate analysis, characteristics related to worse OS included lower Karnofsky, larger extrapontine extension, ring-enhancement, necrosis, non-PR, and increased ring enhancement post-radiotherapy. In the multivariate analysis, Karnofsky, necrosis, extrapontine extension, and therapeutic response can predict OS. A 25% CP reduction (PR) correlated with a 32% volume reduction (R2 = 0.888). Eight patients had discordant therapeutic response classifications according to CP (25%) and volume (32%). This eight patients' median survival time was 13.0 months, significantly higher than that in the non-PR group (8.9 months), in which responses were consistently classified as non-PR based on CP (25%) and volume (32%). We identified correlations between imaging features, therapeutic responses, and OS; this information is crucial for future clinical trials. Tumor volume may represent the DIPG growth pattern more accurately than CP measurement and can be used to evaluate therapeutic response.

It is certainly one of the most feasible ways to extract fresh water from seawater in the face of the current depletion of fresh water resources. Although solar energy as a heat source for desalination is the cleanest and most abundant way, its intermittent and seasonal also poses an obstacle to its practical application. In order to solve the above-mentioned issues, we prepared a series of phase change composites (PCCs) with excellent light-absorbing and magnetic properties by growing MIL-101(Fe) in situ on cotton fabric. All-day desalination through the synergistic action of phase change material (PCM) and magnetic particles. The evaporation rate of PCC can reach 2.76 kg m-2h-1 with an evaporation efficiency of 90.19 % under one sunlight condition. The evaporation rate of sea water under the synergistic effect of magnetic particles and PCM reached 4.53 kg m-2h-1 in the absence of sunlight. This paper provides a new approach to all-day desalination without contact heating.

The severe dependence of traditional phase change materials (PCMs) on the temperature-response and lattice deficiencies in versatility cannot satisfy demand for using such materials in complex application scenarios. Here, we introduced metal ions to induce the self-assembly of MXene nanosheets and achieve their ordered arrangement by combining suction filtration and rapid freezing. Subsequently, a series of MXene/ K+/paraffin wax (PW) phase change composites (PCCs) were obtained via vacuum impregnation in molten PW. The prepared MXene-based PCCs showed versatile applications from macroscale technologies, successfully transforming solar, electric, and magnetic energy into thermal energy stored as latent heat in the PCCs. Moreover, due to the absence of binder in the MXene-based aerogel, MK3@PW exhibits a prime solar-thermal conversion efficiency (98.4%). Notably, MK3@PW can further convert the collected heat energy into electric energy through thermoelectric equipment and realize favorable solar-thermal-electric conversion (producing 206 mV of voltage with light radiation intensity of 200 mw cm-2). An excellent Joule heat performance (reaching 105 °C with an input voltage of 2.5 V) and responsive magnetic-thermal conversion behavior (a charging time of 11.8 s can achieve a thermal insulation effect of 285 s) for contactless thermotherapy were also demonstrated by the MK3@PW. Specifically, as a result of the ordered arrangement of MXene nanosheet self-assembly induced by potassium ions, MK3@PW PCC exhibits a higher electromagnetic shielding efficiency value (57.7 dB) than pure MXene aerogel/PW PCC (29.8 dB) with the same MXene mass. This work presents an opportunity for the multi-scene response and practical application of PCMs that satisfy demand of next-generation multifunctional PCCs.

Developing ultimate electromagnetic interference (EMI) shielding materials that can simultaneously upgrade the quality of generated electricity and the light-thermal-electric conversion efficiency based on traditional thermoelectric devices is crucially desired. Herein, a series of flexible multilayered phase change films (PCFs) is developed by a simple and novel origami strategy. The PCFs are first reported to improve the light-thermal-electric conversion efficiency by as high as 11.3%. Simultaneously, the PCFs could significantly upgrade the generated electricity on average voltage (27.3%), average current (23.8%), and lasting power outputs by 2010 times from microwatts to milliwatts. Besides, the EMI shielding efficiency of PCFs could be tuned from 39.2 to 71.9 dB by the origami process, the wide-range EMI shielding performance could be suitable for varying occasions. Overall, this work provides a promising solution for both the preparation of multifunctional materials, high-efficiency solar energy harvesting and upgrading electricity generation, which shows broad application prospects in EMI shielding, energy storage, and conversion.

This study aimed to investigate the link between COL1A1 production and colorectal carcinoma and assess the value of prognosis and immunotherapy. For this purpose, the transcriptional level of COL1A1 was analyzed. The clinicopathological information and gene expression profile were analyzed to reveal the link between COL1A1 and clinicopathological characteristics. For bioinformatics examination, GSEA and GSVA were utilized. Correlation analysis was implemented to study the causal relationship between COL1A1 and immune checkpoint molecules and inflammation immune cell infiltration. Results showed that in colorectal cancer, COL1A1 was highly expressed and linked with a few clinicopathological characteristics, inflammation and immunological response, tumor immune cell infiltration, and immune checkpoint markers. COL1A1 might likely indicate a bad prognosis and serve as a target of immunotherapy for colon cancer.

Protein PDZ and LIM domain 3 (PDLIM3) is a cytoskeletal protein, colocalizing with α-actinin on the Z line of mature muscle fibers. It plays a key role in dilated cardiomyopathy (DCM), muscular dystrophy, and tumor progression. However, correlations between PDLIM3 expression, prognosis, and tumor-infiltrating immune cells in gastric cancer are unknown. Therefore, we leveraged the Oncomine, GEPIA, GEO, and HPA databases to evaluate PDLIM3 expression in tumors. We also quantified PDLIM3 expression in 15 matched pairs of gastric tumor and nontumor tissues by immunohistochemistry. The Kaplan-Meier method was employed to determine the relationship between PDLIM3 expression and clinical outcomes. GO and KEGG analyses were performed to illuminate the molecular mechanisms of action of PDLIM3. TIMER2.0 and GEPIA were applied to investigate correlations between PDLIM3 expression and gene marker subsets signifying immune infiltration, with TIMER2.0 exploring the correlations between PDLIM3 and related signaling pathways. Gastric cancer tissues were found to express more PDLIM3 than nontumor tissues. PDLIM3 overexpression was associated with shorter OS and PFS of gastric cancer patients (OS HR = 2.02, P = 9.8e - 10; PFS HR = 1.77, P = 7.5e - 06). PDLIM3 was also positively correlated with worse OS and PFS according to gastric cancer staging, Her-2 overexpression, differentiation grade, and Lauren classification. PDLIM3 was shown to be associated with immunological responses by GO, while it was related to PI3K/Akt signal pathways by KEGG analysis. Furthermore, increased PDLIM3 expression was significantly correlated with greater infiltration of CD4+ T cells, CD8+ T cells, macrophages, neutrophils, and dendritic cells. PDLIM3 expression had significant positive correlations with a variety of immune marker subsets. Finally, correlations were found between PDLIM3 and crucial markers of signaling pathways involving PI3K/Akt and p38 MAPK. Thus, upregulation of PDLIM3 was significantly associated with poor prognosis, immune cell infiltration, and activation of two key signal pathways in gastric cancer. We propose that PDLIM3 could be used as a biomarker to predict prognosis and immune cell infiltration in gastric cancer.

With the rapid advance of electronics, the light, flexible, and multifunctional composite films with high electromagnetic interference (EMI) shielding effectiveness and excellent thermal management are highly desirable for next-generation portable and wearable electronic devices. Herein, a series of flexible and ultrathin natural rubber/MXene/carbon nanotubes (NR/MXene/CNTs) composite films with sandwich structure are constructed layer by layer through a facile vacuum-assisted filtration method for EMI shielding and Joule heating application. The fabricated NR/MXene/CNTs-50 composite film, with NR/MXene as inner layer and NR/CNTs as out layers, not only has high EMI shielding efficiency, but also has excellent comprehensive mechanical properties at the thickness of only 200 µm. In addition, the superior environmental durability, high electrothermal conversion efficiency, hydrophobicity, and fine performance stability after periodic cyclic bending make the film possess more value in practical application.

Optically powered nanomotors are advantageous for clean nanotechnology over chemically fuelled nanomotors. The two motor types are further bounded by different physical principles. Despite the gap, we show here that an optically powered DNA bipedal nanomotor is readily created from a high-performing chemically fuelled counterpart by subjecting its fuel to cyclic site-specific optomechanical control - as if the fuel is optically recharged. Optimizing azobenzene-based control of the original nucleotide fuel selects a light-responsive fuel analog that replicates the different binding affinity of the fuel and reaction products. The resultant motor largely retains high-performing features of the original chemical motor, and achieves the highest directional fidelity among reported light-driven DNA nanomotors. This study thus demonstrates a novel strategy for transforming chemical nanomotors to optical ones for clean nanotechnology. The strategy is potentially applicable to many chemical nanomotors with oligomeric fuels like nucleotides, peptides and synthetic polymers, leading to a new class of light-powered nanomotors that are akin to chemical nanomotors and benefit from their generally high efficiency mechanistically. The motor from this study also provides a rare model system for studying the subtle boundary between chemical and optical nanomotors - a topic pertinent to chemomechanical and optomechanical energy conversion at the single-molecule level.

A key capability of molecular motors is sustainable force generation by a single motor copy. Direct force characterization at the single-motor level is still missing for artificial molecular motors, though long reported for their biological counterparts. Here we report single-molecule detection of sustained force-generating motility for an artificial track-walking molecular motor capable of autonomous chemically fueled operation. A single motor plus its track (both made of deoxyribonucleic acids or DNA) is assembled, operated and detected under magnetic tweezers by a method designed to overcome difficulty from the motor's soft double-stranded track. The motor shows self-directed walking by ∼16 nm steps up to a distance of 120 nm (covering the entire track), yielding a stall force of ∼2-3 pN. These results imply a reasonably efficient chemomechanical conversion of the motor compared to a high-efficiency biomotor. The stall force is near the level of translational biomotors powering human muscles and allows similar force-demanding applications by their artificial counterparts. This single-motor study reveals fast subsecond steps, suggesting big room for improvement in the speed of DNA motors in general. Besides, the established single-molecule method is applicable to force measurements of many other DNA motors with soft tracks.

Cullin 7 is the 'scaffold' of the cullin-RING-based E3 ligases which catalyze the final step of the ubiquitination cascade in eukaryotic organisms. Although one study has reported the involvement of cullin 7 in the invasion and migration of breast cancer cells without further exploration of its mechanisms, the expression of cullin 7 in breast cancer tissues and its clinical significance have not been reported. The present study evaluated cullin 7 protein expression in malignant and benign breast tissues using immunochemistry, and further analyzed the association of positive cullin 7 expression in breast cancer tissues with clinicopathological characteristics of breast cancer patients. Cullin 7 expression was further silenced in breast cancer cells by siRNA and its functions and mechanisms in cell proliferation and invasion were investigated. The results showed that high cullin 7 expression was significantly correlated with pathological stage (P=0.013) and lymph node metastasis (P=0.022) of breast cancer. Patients with high cullin 7 expression had a shorter overall survival rate than those with low cullin 7 expression (P=0.037). Silencing of cullin 7 expression significantly inhibited the proliferation (P<0.05) and invasion (P<0.05), increased S1 phase (P<0.05), but decreased G2 phase (P<0.05) in MDA-MB-231 and BT549 cells. In addition, silencing of cullin 7 expression decreased cyclin A, but increased p21 protein expression. Moreover, silencing of cullin 7 expression altered the cell shape, caused disorder in microtubules, and increased the microtubule regeneration in MDA-MB-231 and BT549 cells. In conclusion, cullin 7 is overexpressed in breast cancer tissues which is associated with the development and prognosis of breast cancer. Cullin 7 is involved in the proliferation and invasion of breast cancer cells by regulating the cell cycle and microtubule stability.

A modified purification procedure is described for effectively eliminating dead cells after hepatocyte cryopreservation. Isolated hepatocytes from six pig tissue samples were cryopreserved in liquid nitrogen for 2 weeks. After thawing, we developed a pre-incubation step prior to gradient centrifugation. The hepatocytes were subsequent cultured in suspension overnight (12-16 h), and then dead cells were eliminated by Ficoll 400 purification. The results showed that a high viability (mean of 96%) of cells was obtained, with a low viable cell loss in number (2-5%), by using this modified method.
